Rationale/ Salient Characteristics
For all items:
• All reagents must be easily scalable and highly sensitive
• Engineered for live cell imaging
• Optimized for multi-well plates and automation equipment
• Easily scalable
• Reagent kits and assays must be optimized for Primary high-throughput screening, secondary screening, and lead optimization.

Rabbit Reticulocyte Lysate, Nuclease-Treated Kits, 30 reactions:
• Prepared from white rabbits using a standard protocol (1) that ensures reliable and consistent reticulocyte production in each lot.
• Reticulocytes purified to remove contaminating cells, which could otherwise alter the translational properties of the final extract.
• Treated with micrococcal nuclease to destroy endogenous
• Lysate contains the cellular components necessary for protein synthesis (tRNA, ribosomes, amino acids, initiation, elongation and termination factors).
• Optimized further for mRNA translation by the addition of the following:
o Energy-generating system consisting of prequalified phosphocreatine and phosphocreatine kinase
o Mixture of tRNAs to expand the range of mRNAs that can be translated
o Hemin, to prevent inhibition of initiation
o Potassium acetate and magnesium acetate
• Provided in 200μl aliquots. Each system contains sufficient reagents to perform approximately 30 translation reactions per milliliter of lysate. Includes:
o 1ml Rabbit Reticulocyte Lysate, Nuclease Treated (5 × 200μl)
o 50μl Amino Acid Mixture Minus Methionine, 1mM
o 50μl Amino Acid Mixture Minus Leucine, 1mM
o 50μl Amino Acid Mixture Minus Cysteine, 1mM
o 10μl Luciferase Control RNA

Luciferase Control RNA, 20 ug:
• Safe, non-radioactive format to monitor control activity
• Supplied at a concentration of 1mg/mlKjk
• uncapped in vitro-transcribed RNA containing a 30-base poly(A) tail that produces functional luciferase when translated.
• Control reactions are monitored easily by a luciferase assay for the production of luminescence generated from the full-length luciferase.
• Control activity can be monitored without the use of radioactive materials.

ONE-Glo Luciferase Assay System, 100 ml:
• provides a highly sensitive, robust, homogeneous assay for detection of firefly luciferase reporter gene expression in mammalian cells.
• Ideally suited for high- and ultrahigh-throughput applications.
• Room temperature or 4°C storage options.
• Ideal for use in high-density (384- and 1536-well) microplates.
• Brighter, Longer-Lasting Signal. Optimized for batch and continuous-process handling, the extended bright light output allows high sensitivity, especially for extended incubations.
• Reduced Unwanted Effects from Sample Components. Less sensitive to culture media, phenol red and luciferase inhibitors than other luciferase assays.
