Only 10xGenomics is capable of providing the necessary kits needed for the Government because the kits are proprietary to 10xGenomics and they are the original manufacturer. The material or service must be compatible in all aspects (form, fit, and function) with existing systems presently installed. The current equipment is the 10x Chromium™ system, and the new item/service must coordinate, connect, or interface with the existing system by being recognized by the 10x Chromium™ system and, through the operation of the 10x Chromium™ system, produce an emulsion of cells, gel-beads and reagents compatible with single-cell chemistry. Therefore, only this source can furnish the requirements, to the exclusion of other sources.
Background/Description of Requirement

PURCHASE DESCRIPTION

 

 

  1. Statement of Need and Purpose: The Spinal Circuits and Plasticity Unit needs reagents for single-cell RNA-sequencing + ATAC-seq in order to simultaneously profile the transcripts and open chromatin of embryonic spinal cord neurons to further resolve the full breadth of embryonic spinal cord neuron types.
     
  2. Background Information and Objective: The National Institute of Neurological Disorders and Stroke (NINDS) Spinal Circuits and Plasticity Unit conducts frequent single-cell transcriptomics work in order to continue to develop the atlas of spinal cord neurons which is currently the standard reference for the field. We have recently developed an embryonic transcriptomic atlas of the mouse spinal cord, which is currently the largest and most comprehensive yet produced. We seek to complement this atlas with a corresponding map of open chromatin in order to provide us and the field with a reference for gene regulatory networks in the spinal cord. We thus need reagents allowing microfluidic collection and sorting of spinal cord nuclei, production of both cDNA and DNA derived from open chromatin, as well as production of sequencing-ready DNA libraries from open chromatin-derived DNA and cDNA respectively.
