Single-Sole Source Determination
During market research, it was found that GenScript USA, Inc is the only vendor that can provide the required product. It was found that no similar product could duplicate what their method due to their proprietary tagging approaches and GenScript’s vast index of molecules. This is in addition to their expertise in DEL screening which is important for this acquisition. Therefore, with these facts in mind, it was found that Magstim Inc. is the only vendor that can provide the needed product.
Background/Description of Requirement

The National Center for Advancing Translational Sciences (NCATS) is a part of the National Institutes of Health (NIH), with a mission to catalyze the generation of innovative methods and technologies that will enhance the development, testing, and implementation of diagnostics and therapeutics across a wide range of human diseases and conditions. The Early Translational Branch (ETB) is tasked with identifying and developing novel small molecule leads against a wide spectrum of human disease targets, which it accomplishes by screening thousands of small molecules in collaboration with both intra-and extramural researchers.

In pursuit of this goal, discovering small molecules that could serve as starting points for therapeutics is of great interest within NCATS. Medicinal chemistry optimization is often an important exercise in our development of small molecules to test the therapeutic hypothesis in disease models. NCATS uses several strategies and modalities to address many aspects of design approaches to discover novel small molecules.

One such strategy is the use of DNA-encoded chemical libraries (DELs) which represent a versatile platform for the discovery of small-molecule ligands to protein targets of biological interest. DELs are collections of molecules, individually coupled to distinctive DNA tags serving as amplifiable identification barcodes. Thanks to advances in DNA-compatible reactions, selection methodologies, next-generation sequencing, and data analysis, DEL technology allows the construction and screening of libraries of unprecedented size, which has led to the discovery of highly potent ligands, some of which have progressed to clinical trials.

This requisition is to acquire a DEL chemical library kit, which will allow NCATS to find more suitable drug leads for our target disease models we are studying.

The compound library that is being assembled needs to ensure that services include:

- Small molecules must have validated biological and pharmacological activities.

- Must be suitable for high-throughput screening (HTS) and high-content screening (HCS).

- Must come with detailed biological and chemical information with LC/MS and NMR reports to ensure high quality.

- Guaranteed to have a purity of >95% (accuracy +/-5%) that is to be confirmed by NMR and/or LCMS spectra.

Delivery is expected within 3-12 months after receipt of order.
