This acquisition is conducted under the authority of the Federal Acquisition Regulation (FAR) Part 13—Simplified Acquisition Procedures, Subpart 13.106-1 (b) (1), Soliciting from a single source and is not expected to exceed the simplified acquisition threshold. Contracts awarded using FAR Part 13—Simplified Acquisition Procedures and at a dollar value less than the
simplified acquisition threshold are exempt from the requirements of FAR Part 6—Competition Requirements.

Hitech Instruments, Inc is the only authorized provider of this instrument at the NIH. Hitech Instruments is Zeiss’ exclusive distributor of this product at NIH and as such is the only authorized source to purchase from.

Hitech Instruments, Inc. has provided a sole source letter to the Government certifying that no other company is authorized distributor of Hitech Instruments, Inc. brand products.



Background/Description of Requirement

Acquisition Authority: This acquisition is being conducted under Federal Acquisition Regulations (FAR) Part 13 – Simplified Acquisition Procedures, and the resultant contract award will include all applicable provisions and clauses in effect through Federal Acquisition Circular 2021-06 published in the Federal Register on July 12, 2021.

A Firm Fixed Price (FFP) Purchase Order award not exceeding the Simplified Acquisition Threshold is anticipated from any contract resulting from this requirement.

The Transcription Systems Dynamics and Biology Uni (TSDBU), Laboratory of Molecular Biology and Immunology (LMBI) with the National Institute on Aging requires the use of ZEISS Axio Vert.A1 FL-LED microscope system to allow for monitoring fluorescence in DAPI, GFP, TxRed, and Cy5 channels and capturing the fluorescence from the culture dishes.  This microscope is suitable for quick checking the fluorescence regularly needed in the tissue culture room.

The transcription factor NF-κB regulates multiple aspects of innate and adaptive immune functions and serves as a pivotal mediator of inflammatory responses. To monitor the endogenous NF-κB signaling in innate immune cells and the role of non-coding genomic regions around NF-κB regulated genes, LMBI TSDBU has generated immortalized macrophages from CRISPR Knock-in fluorescent mouse model.  LMBI TSDBU will generate mutants containing genomic deletions in different non-coding regions and monitor their effects on NF-κB signaling dynamics. For sorting the mutant cells, LMBI TSDBU will use fluorescent dCas9-gRNA RNP complex which needs to monitor regularly in the microscope to optimize the culture and sorting conditions. To regularly monitor the fluorescence of these immortalized wild-type and mutant cells, a fluorescence tissue culture microscope is needed in the tissue culture room.
