The AGNCU needs to do a complete characterization of the metabolic profile of Drosophila melanogaster to determine how physiology changes with aging, and how those changes are affected by the environment, specifically the presence or absence of bacteria.
Background/Description of Requirement

In the process of investigating a Drosophila model of human neurodegenerative disease, the AGNCU discovered that removing bacteria from the environment completely rescues the lifespan of individuals carrying mutations that cause neurodegeneration. This makes it essential to understand in detail how the presence or absence of bacteria alters the state of the organism itself. Previous experiments have investigated the effect this has on gene expression, but we need to connect those changes in gene expression with their eventual effects on the direct physiology of the animal. The best way to do this is by characterizing the complete metabolic profile of the animal under the same conditions that were used for gene expression analysis. NOTE: The key requirement for this project is identification of a large enough set of metabolic components to allow statistically significant identification of metabolic pathways that are altered by the perturbations under study. This requires maximum efficiency of separating different constituent chemicals and the largest possible database for identifying chemcial species generated by mass spectrometry. The greatest weakness in metabolomic studies is the inability to identify the precise chemical nature of compounds that are flagged as significantly altered in level.
