Rationale/ Salient Characteristics
The essential characteristics of the transgenic genotyping service that limit the availability to a sole source are daily pickup and transportation of samples from the NIH Bethesda campus, plus storage of remaining samples after analysis is completed, and return of samples to the Laboratory on Neuroplasticity as needed. Only this suggested source can furnish these requirements, to the exclusion of other sources, because other companies do not offer any of these options.
Single-Sole Source Determination
Transnetyx is the only contractor on NIH BPA Program List that provides sequencing services that can genotype many custom and non-standard mouse lines and the reliability of the data is accurate. The essential characteristics of the transgenic genotyping service that limit the availability to a sole source are daily pickup and transportation of samples from the NIH Bethesda campus, plus storage of remaining samples after analysis is completed, and return of samples to the Cellular Neurophysiology Section as needed. Only this suggested source can furnish these requirements, to the exclusion of other sources, because other companies do not offer any of these options.
Background/Description of Requirement

Research in the Cellular Neurophysiology Section focuses on circuit and cellular properties of dopaminergic neurons located in the midbrain. We are interested in the physiology of a subset of dopamine neuron located in the substantia nigra pars compacta (SNc) that undergo selective cell death in Parkinson’s patients.  Specifically, we are interested in understanding how synaptic and intrinsic conductances differ among subpopulations dopamine neurons with the hope that this may provide insight to why SNc dopamine neurons are particularly vulnerable in Parkinson’s disease.  Our goal is to identify novel physiological and genetic markers in single SNc dopamine neurons which may be useful in defining subpopulations of these neurons in a meaningful way.

To identify different dopamine neuron subpopulations, we use genetically labeled mice that express fluorescent reporters. In other experiments, we use a variety of transgenic mice to study different aspects of dopamine neuron physiology. In order to perform these experiments, therefore, an urgent and basic need for the lab is to have genotyping information available for the different mouse lines in our mouse colony.

As part of the genotyping service, our laboratory will ship small tissue samples from each mouse along with the name of the gene that we would like to identify. After receiving the tissue samples, Contractor will use an automated genotyping system to identify the gene of interest within the genetic materials that we provide. The company also gives us access to software that will help organize our colony.

Deliveries:

There will be no visits taking place. Instead, samples will be shipped to contractor. We require that contractor genotype our tissue samples and allow us to use their free colony management software. Results are provided within 72 hours of samples submission.
