Rationale/ Salient Characteristics
To fulfill the NINDS BMMCU’s requirement, the fermenter/bioreactor shall be brand-name or equal to the Sartorius A MO UniVessel Glass fermenter/bioreactor (“Equipment”) and meet the following salient physical, functional, or performance characteristics:

a. One Biostat A Control Tower (table top configuration) or equal: includes control of stirred tank, motor drive, three integrated fast load pumps with fixed speed (on/off control), integrated amplifier for pH, dissolved oxygen (DO), temperature, foam/ level, power cord, two external signal inputs (4 to 20 mA) for connecting external measurement systems, measured signal display (range 0 – 100%), USB port, ethernet port, and installation kit

b. One Biostat A operating software or equal for laptop/PC: includes functions to control stirrer speed, temperature, pH, DO, foam/level. Also includes data parameters storage and export functions, probe calibration function, and alarm functions

c. One Biostat A aeration system for microbial fermentation or equal: includes continuous and automatic aeration control for air and O2, two gas flow control units - 1 per gas segment, flow rates of the gas flow control units: Air | O2: 100-7500 ccm, automatic gas flow control via DO controller

d. One Biostat A measurement package or equal: includes one pH probe with combination electrode, digital communication with control tower and connection cable; one temperature sensor integrated into pH probe; one ampere-based DO probe with digital communication with control tower and connection cable; (d) one foam/level probe with connection cable

e. One integrated pH monitoring and pH control system: includes proportional-integral-derivative (PID)-controlled dosing of base via integrated fast load pumps

f. One integrated DO monitoring system and controller for PID-controlled infusion of oxygen and impeller rate: includes
automatic DO control and parallel control of up to 4 actuators, with individual control of parameters for each actuator

g. One integrated anti-foam detection system: combined anti-foam level detection and control

h. Integrated cooling system: includes one external recirculating chiller (approx. dimensions: 220 (w) x 490 (h) x 340 mm (d), cooling capacity: 150 W), one cooling finger, one hose set for external recirculating chiller, connections for cooling finger, connections for exhaust air cooler and external power supply.

i. One heating jacket powered by control tower

j. One exhaust air cooling system for Biostat UniVessel Glass 2L or equal: includes exhaust air cooler for installation in 19 mm hot plate port, quick coupling halves for cooling water inlet and outlet. Material in contact with medium: Stainless steel AISI 316L.

k. One substrate pump control of external, variable speed pump

l. One external, variable speed pump with connection cable to fermenter/bioreactor

m. One Biostat A UniVessel 2L, single wall or equal: includes polished stainless steel lids, stainless steel stand to support culture vessel. Material in contact with medium: stainless steel AISI 316L and borosilicate, EPDM (FDA). Must have separate 6 mm ports for foam/level probe, sparger, baffles, inoculation port, and harvest pipe, and four reserve ports. Must have separate 19 mm ports for exhaust cooler. Must have 4-way port with hose nozzles for tubing, ID = 1.6 mm. Must have cooling finger port. Must have separate, 12 mm ports for DO and pH probes.

n. One standard coupling: includes coupling between stirrer shaft and motor and rubber sleeve for direct drive

o. One Biostat UniVessel Glass 2L or equal stirrer with single mechanical seal

p. Two 6-blade disk impellers, stirrer diameter 53 mm

q. Four baffles for UniVessel Glass 2L or equal

Background/Description of Requirement

This is a notice of solicitation in accordance with FAR 5.202(a)(2).

The solicitation includes attachments, the inclusion of which is restricted by this electronic bid board. An e-copy of the solicitation and all attachments is available by request to the above cited contract specialist.

The National Institute of Neurological Disorders and Stroke (NINDS), Brain MRI Molecular Contrast Unit (BMMCU) requires a fermenter/bioreactor that is brand name or equal to the Sartorius Biostat A MO UniVessel Glass Fermenter/Bioreactor for the production of large-scale protein yield.

The National Institute of Neurological Disorders and Stroke’s (NINDS) Brain MRI Molecular Contrast Unit (BMMCU), led by Dr. Alan Koretsky, is developing a novel class of brain MRI contrast agents which will provide improved contrast of specific brain pathologies.  Achieving sufficient amounts of contrast agent requires scaling up production of recombinant proteins to a level above the typical yield producible through standard cultivation methods utilized by the BMMCU Laboratory. Therefore, the BMMCU requires a Sartorius A MO UniVessel Glass or equal fermenter/bioreactor to enable it to produce sufficient protein yield for development of the novel class of MRI contrast agents.

Interested parties may identify in writing their interest and capability in response to this requirement. Responses to this notice shall contain sufficient information to establish the interested parties’ bona-fide capabilities for fulfilling the requirement and include: unit price, list price, shipping and handling costs, the delivery period after contract award, the prompt payment discount terms, the F.O.B. Point (Destination or Origin), the Dun & Bradstreet Number (DUNS), the Taxpayer Identification Number (TIN), and the certification of business size. All offerors must have an active registration in the System for Award Management (SAM) www.sam.gov.
