Background/Description of Requirement

The National Institute on Aging's (NIA) Laboratory of Neurogenetics (LNG) utilizes Nanopore sequencing technology, which has been in use for several years and has become optimized for use in in-house sequencing.  This technology offers direct sequencing of native DNA and RNA as well as amplified samples in real-time.  LNG has purchased the sequencing equipment so that samples can easily be run in flow cells.  These flow cells have the advantage of reading any length nucleic acid and can be used until the nanopores have reach their life expectancy.  New flow cells must therefore be purchased as the used ones become inadequate.

New flow cells and direct RNA sequencing kits supplies are required to perform direct RNA sequencing (dRNA-Seq) for profiling the full-length transcriptome of mouse tissue and human cell lines. Optimization will initially be performed in HeLa and HEK293T cells. This will allow LNG to then deep sequence the aged-mouse hippocampus in WT and APPPS1, an Alzheimers disease model system in which Amyloid precursor protein and Presenilin genes are inactivated (a double KO mice) as well as control and TDP-43 KD human iPSC derived neurons, which serves as a model system for Amyotropic Lateral Sclerosis and Frontotemporal Dementia.
