Description of Requirement:
The over expression of adenylyl cyclase in the mouse heart produces a strong beating heart with the upregulation of many pathways. The 14-3-3 protein is one of these proteins that emerge as central for the heart survival and regulation of the cell cycle and mitosis check points.
Here we plan to analyze the role of the 14-3-3 protein by detecting its binding partners in the AdCy8 over expression model. This requirement is part of an ongoing project in which initial analysis was done on tissues from the HAC8 project in 2018 and 2019. This analysis will be done through the use of a special method called immunoprecipitation (IP) followed by mass spectrometry analysis (MS) and comparison of the results with the wild type mouse model.
Specific Requirements:
- Perform Immuno Precipitation (IP) tests in control and AC8TG tissues. Method development and testing of pan 14-3-3 on eight (8) samples. The immune precipitation requires the use of a specific antibody against the 14-3-3 protein. This antibody needs to recognize every member of the 14-3-3 protein and be suitable for immono-precipitation. Mass spectroscopy run for the detection and the recognition of the 14-3-3 protein binding partners.
- Perform 14-3-3 IP analysis on the eight (8) samples.
- Analyze and compare the data results.
Delivery or Deliverables:
The Contractor will provide a report, in the form of an Excel spreadsheet, of the results from each step of the work required and as described in the Scope of Work.

Contract Type:
A Fixed-Price type of Purchase Order award not exceeding the Simplified Acquisition Threshold is anticipated for any contract resulting from this requirement. The anticipated award date is January 05, 2021.
