Acquisition Purpose and Objectives:
This acquisition is necessary to provide a stage top chamber for an existing microscope to the iPSC Neurodegenerative Diseases Initiative (INDI) Project within the new Center for Alzheimer’s Disease and Related Dementias.
Description of the Item or Service:
Stage Top microscope Chamber is to provide a stable environment for iPS cells to survive during long acquisition times on the microscope. The cells need certain temperature, humidity and CO2 level in order to survive. If they do not have the support of the chamber, the cells will not survive which will render the data collection invalid.
Salient Characteristics:
a. Temperature Range: from 3°C above ambient to 60°C
b. Temperature Accuracy: ± 0.1°C in sample feedback mode, ± 0.3°C in chamber feedback mode
c. Compatible with: Bold Line T, UNO Combined and H401-T Controllers
d. Chambers available for any XY stage / piezo insert
e. Embedded temperature sensor in heated glass lid and in chamber body
f. Interchangeable magnetic inserts allow to host Petri, Slides and MW Plates
g. Magnetic locks hold the Petri, the Slides and the MW Plates in the correct position inside the chamber
h. Perfusion holes available for inlet and outlet of tubes
i. Sliding lid allows easy access to the sample
j. Optional lids are available for Koehler Illumination, laser safety and injection during imaging
Manufacturer warranty for one calendar year beginning date of receipt.

Project Background:
The NIA Center for Alzheimer's Disease and Related Dementias (card) is working to create a national resource of isogenic sets of Alzheimer’s Disease and Related Dementias mutated iPSC lines, complimented by foundational phenotypic data sets from differentiated disease-relevant cells derived from these lines. We are working to create a national resource of isogenic sets of Alzheimer’s Disease and Related Dementias mutated iPSC lines, complimented by foundational phenotypic data sets from differentiated disease-relevant cells derived from these lines. This stage top chamber furthers the functionality of the Biopipeline microscope, allows for long term cell imaging while maintaining environmental stability. This is essential in keeping the cells alive especially when doing extended acquisitions with this microscope. The iPS cell lines are essential in studying the pathogenesis of neurodegenerative these diseases.
