10X Genomics single-cell analysis reagents listed herein are available from only one source, and competition is precluded for the reasons indicated below. There are no substitutes available.
Background/Description of Requirement
The essential characteristics of the 10X Genomics Chromium single-cell “Drop-Seq” kit that restrict the availability to a sole source are: The Drop-Seq technique is a newly developed and patented technique by which a flow of beads suspended in lysis buffer and a flow of a single-cell suspension are brought together in a microfluidic chip that generates nanoliter-sized emulsion droplets, which are then placed on a microfluidics chip and analyzed by the Chromium Controller machine. The Controller machine already exists and is available for our use. This machine allows for the high-throughput and rapid processing of the sample. It will process only 10X Genomics microfluidic chips, which cannot be obtained from any other vendor. A key advantage of the protocol is that a high number of single-cell RNA-seq libraries can be generated at low cost. The Chromium Single Cell 3’ Reagent Kit enables deep profiling of complex cell populations with high-throughput 3’ digital gene expression analysis of individual cells. Each bead contains a unique barcode so that constructed RNA library can be immediately coupled to any Illumina-based sequencing system, unlike some in-house versions of single-cell sequencing. • The methodology recovers up to 65% of all loaded cells—which is the best recovery rate currently available for Drop-seq kits. This is important for identifying rare cell populations • Low rate (0.9% for 1000 cells) of cell doublet formation in their patented microfluidics chamber-based droplet encapsulation. Improves confidence that you are looking at the RNA from a single cell, again important for looking at rare cell populations • Speed is faster than any other platform on the market, which eliminates bias over time. i.e., a cell sorting session could take hours, and there will be changes over time in the RNA quality for cells collected at the beginning of the session vs. those collected at the end. • This is the only company that sells reagents enabling VDJ transcript analysis for B and T cells (i.e., the ‘variable’ regions of the genome leading to adaptive immunity in T and B cells). Single-cell sequencing provides a number of advantages, including: • Often masked by bulk average measurements, single-cell sequencing can detect biologically relevant gene expression signals from up to 100,000 individual cells per chip across heterogeneous cell types. • Rare or novel cell types may also be uncovered and characterized based on environmental or experimental conditions. .
