Single cell RNA sequencing allows for the identification of mRNA transcripts from single, dissociated, cells. This information is then used to elucidate the funcition of the tissues from which the cells originated. In order to process samples, the sequencing platform requires use of certain consumable reagents. This order is for enough of these consumables in order to run 16 sequencing reactions. This is a new requirement. : Chromium Next GEM Chip G single cell kits (#1000120) and Library & Gel Bead Kit (#1000121) from 10x Genomics.

The reagents listed herein are available from only one source. There are no substitutes available.
Rationale/ Salient Characteristics
Reagents must be compatible with the 10x Genomics Chromium Controller instrument operated by the NICHD genomics core facility. Reagents must allow for the preparation of the genomic library and allow for gel bead tagging and microfluidic separation of single cells.
Single-Sole Source Determination
The essential characteristics of the reagents that limit the availability to a sole source are the ability to be compatible with the 10x Genomics Chromium Controller instrument operated by the NICHD genomics core facility. Reagents must allow for the preparation of the genomic library and allow for gel bead tagging and microfluidic separation of single cells.

The material or service must be compatible in all aspects (form, fit, and function) with existing systems presently installed. The current equipment is a 10x Genomics Chromium Controller sequencing instrument, and the new item/service must coordinate, connect, or interface with the existing system by integrating into the current instrument and be compatible with the microfluidic pathway allowing single cells to be combined with the included beads and allow generation of the single cell RNA sequence.

The new items are "direct replacement" parts / components for existing equipment.
Background/Description of Requirement
Single cell RNA sequencing allows for the identification of mRNA transcripts from single, dissociated, cells. This information is then used to elucidate the function of the tissues from which the cells originated.
