Single-Sole Source Determination
These projects are designed to identify the transcriptomic response to Allopregnanolone in various regions of the mouse brain. Allopregnanolone is known to be anxiolytic and is thought to play a key role in reproductive mood disorders such as postpartum depression (PPD) and premenstrual dysphoric disorder (PMDD). The effects of ALLO on GABAAR subunit gene expression will be measured first in isolation in ovariectomized mice (where the natural cycling of ovarian steroids across the estrus cycle has been eliminated), then within the context of the estrus cycle.

The items requested are reagents are sufficient to perform an RNA-seq experiment with a sufficient N for a final analysis (n=6 per group) across 6 conditions: post-OVX + vehicle, post-OVX + ALLO, proestrus, estrus, metestrus, late diestrus. With these reagents we’ll able to sequence mouse brain regions implicated in mood and behavior (including the Striatum and Amygdala) of mice after administration of allopregnanolone or vehicle and across the estrus cycle: (n=6 in each condition (6) and brain region (2), for 72 total samples).

As a supplement in this acquisition package, the Life Technologies (part of ThermoFisher)  has provided a company statement stating that they are the sole manufacturer of the Ion GeneStudio™ S5 Series of sequencing instruments, the Ion Proton™ System, the Ion One Touch™ 2 System, and the Ion Chef ™ Instrument and their associated consumables including Ion library preparation, template, sequencing, and barcode reagent kits as well as semiconductor chips. Ion sequencing represents a unique technology, using semiconductor chips to directly measure pH changes that occur in natural sequencing reactions.
