Purchase Description: MIMMS2.2 Head Unit For Multiphoton Microscopy System

Salient characteristics:

Detection arm module
• Redesign of the detection arm to accommodate a new piezo based objective scanner from PI (the old one is now obsolete), the piezo objective scanner from Thorlabs, as well as the stationary objective option (no fast-z scanner).
• Complete redesign of the bottom part of the detection arm to make it stiffer and improve stability for the applications where an objective lens needs to be fast scanned through the full stroke at the maximum achievable scan rates.
• ROI RGG laser scanner:
Complete redesign of the galvo-galvo submodule to accommodate for the ScannerMax galvos. In addition, miniature XY translation stages were added to facilitate precise alignment of the scanned laser beam onto the optical axis of the microscope.
Widefield/1p-epifluorescence and DMD-based opto-stimulation modules:
• Redesigned both to combine them together at the same port. This frees up a port for the second laser scanner module, which is becoming a popular option. In addition, this facilitates an option of widefield imaging with per pixel control of illumination intensity. This can help achieving uniform illumination for imaging of voltage indicators, for example. Refer to MIMMS 2.1 documentation if the DMD option is not needed for the Widefield/1p-epifluorescence module.
Custom Electronics:
• Redesigned mechanisms for moving the primary dichroic mirror, widefield module mirror and the auxiliary port mirror.
• Updated Galvo 2U controller CAD model for ScannerMax galvo controller PCB.
• Added CAD models for the PMT controller and the resonant mirror controller.

The essential characteristics of the customizable MIMMS2.2 that limit the availability to a
sole source are this is the only available commercial offering full compatibility with
existing equipment. Only this suggested source can furnish the requirements, to the
exclusion of other sources, because as market research has shown SuperPostdoc, LLC
is the only commercially available system meeting set specified standards. A The
complete component system meets all the technical requirements as listed in the
product description salient requirements as reproduced below
Background/Description of Requirement

The UNCB studies the function of cortical network activity in vivo using single-cell optogenetic stimulation. A single stimulus or motor action changes the activity of many cells in the cerebral cortex, and it is thought that patterns of activity across many cells are the fundamental unit of cortical function. However, we have been unable to understand how the brain uses activity patterns across multiple cells, principally because, as a field, we have been unable to change the activity of multiple individual cells in a patterned way. Two-photon optogenetic stimulation can provide such control, and thus has enormous promise to reveal how the brain works. Several groups worldwide, including our group, are working to deploy this approach in behaving animals. To do this, we develop custom optical microscopes which are able to both image and stimulate neurons using highpulse power lasers. Currently, the UNCB has MIMMS (Modular In vivo Multiphoton Microscopy System)2.1 head that was purchased in 2020. The Bill of Materials was updated to replace all the components that became obsolete or prohibitively expensive since 2020.
