Single-Sole Source Determination
The objective of this purchase is to have a stock of cell culture reagents required for the culture of iPSC and generation of iMGL. Ultimately, this will facilitate the characterization of iMGL derived from individuals with Down Syndrome (DSWR), Down Syndrome Regression Disorder (DSRD), and karyotypically normal (KN) controls. For iPSC culture, mTeSR Plus media is utilized for long-term, quality culture of iPSC. Due to the challenge of maintaining cells in a pluripotent state, very particular medias have been developed to minimize risk of expansion of cells that have developed genetic abnormalities that are advantageous for growth. The use of iPSC for culture is essential in the field of neuroscience, due to the challenge of accessing neural cell types of interest from many patient populations. Down Syndrome Regression Disorder is a disorder that typically occurs between 15-25 years of age, and is fortunately not terminal. Because of this, there is little to no access to primary microglia from individuals with DSRD. Additionally, there is currently no animal model of the disorder. Thus, it is necessary to use cell models to elucidate cell-autonomous contributions to the disorder.

To specifically study microglia, there are multiple stages of the differentiation process. The iPSC will first be differentiated to a hematopoietic progenitor cell, which is similar to a hematopoietic stem cell; a cell type capable of myelopoiesis. This first stage requires the iHPC media kit, and is comprised of multiple stages itself. The second step of differentiation is the microglial differentiation, a process which is divided amongst two separate kits: the iMGL Differentiation kit and the iMGL maturation kit. Upon terminal differentiation of these cells, we complete several functional assays to compare across the DSWR group, DSRD group, and KN control group.
Background/Description of Requirement
