Background/Description of Requirement

The National Institutes of Health (NIH) is the nation’s leading medical research agency and the primary Federal agency whose mission is to seek fundamental knowledge about the nature and behavior of living systems and the application of that knowledge to enhance health, lengthen life, and reduce illness and disability, conducting, supporting and making medical discoveries that improve people’s health and save lives.

Drug development methodologies currently followed are often time consuming, expensive, and highly inefficient. This is mainly due to a lack of physiologically relevant model systems exemplified by decades of reliance on non-human animal models for disease modeling and drug discovery. Recent progress in stem cell biology, specifically reprogramming of somatic cells to induced pluripotent stem (iPS) cells and their subsequent directed differentiation to different cell types, including but not limited to cells  neural lineages, has provided physiologically relevant systems for modeling human diseases and drug discovery studies.  The aim of this project is to systematically identify gene expression changes at the single cell level upon treatment of iPSC-derived Astrocytes with reactive state inducers: Interleukin-1, TNF-Alpha, and C1Q.
