Statement of Need and Purpose:
The purpose of this acquisition is to obtain one (1) x KAPA mRNA Capture Kit only (96rxn), two (2) x KAPA Pure Beads 60ml, and four (4) x KAPA mRNA Hyper Prep (96rxn), and one (1) x KAPA RNA Hyper+RiboErase HMR (96rxn) which are required to continue to perform transcriptomic analysis by the Stem Cell Translation Laboratory (SCTL). The reagents allows low input RNA for RNAseq library preparation and the procedure was established and compatible with current automation platform in SCTL.

The material must be compatible in all aspects (form, fit, and function) with existing systems presently installed and must coordinate, connect, or interface with the existing equipment or system. The KAPA Hyper reactions are automated in SCTL existing automation system (Beckman i7) to ensure high throughout and high accuracy sequencing library preparation.
Background/Description of Requirement

Background Information and Objective: 

The SCTL requires the acquisition of one (1) x KAPA mRNA Capture Kit only (96rxn), two (2) x KAPA Pure Beads 60ml, and four (4) x KAPA mRNA Hyper Prep (96rxn), and one (1) x KAPA RNA Hyper+RiboErase HMR (96rxn) to enable diverse library preparation methods for a variety of sample types and application needs. 

The KAPA mRNA HyperPrep Kit utilizes novel chemistry that enables the combination of enzymatic steps and fewer reaction purifications, resulting in a truly streamlined solution for the preparation of high-quality RNA-seq libraries. The strand-specific workflow is flexible—supporting library construction from lower-input amounts and degraded samples and mRNA capture. Kits contain all reagents required for RNA enrichment (if performed) and library preparation. Reaction buffers are supplied in convenient formats comprising all of the required reaction components. This minimizes the risk of RNase contamination, ensures consistent and homogenous reaction composition, and improves uniformity among replicate samples. Similarly, a single enzyme mixture is provided for each step of the library construction process, reducing the number of pipetting steps. The KAPA RNA HyperPrep Kit with RiboErase (HMR) for Illumina sequencing contains all of the buffers and enzymes required for depletion of ribosomal RNA (rRNA) and the rapid construction of stranded RNA-Seq libraries from 25 ng – 1 µg of purified total RNA. The procedures are automated in SCTL lab existing liquid handler, the protocols are provided and validated by Roche automation team. The KAPA mRNA Capture Kit only (96rxn), and KAPA Pure Beads 60ml are required for reagent overage volume on automation.
