- A Nanodrop device for analyzing DNA and RNA.
The purpose of this equipment is to analyze quantity and quality of DNA and RNA in solution using only a small volume (~1uL).
This equipment will be critical for the central mission of the Glial Cell Biology unit to study the cell biology of oligodendrocytes. We are requesting this item to analyze DNA constructs used for electroporation into cells and DNA/RNAs as part of workflows for qPCR and RNA-seq experiments.
1. Is a standalone instrument with local control and a high resolution touchscreen interface. Touchscreen display accommodates front to back tilt and left to right slide adjustments.
2. Uses a patented (US patents 6,628,382 and 6,809,826) sample retention system that enables direct measurements of sample loaded directly onto a polished stainless steel pedestal and allows for easy cleaning with no carryover using a laboratory wipe.
4. Measures 1-2 μL of high- and low-concentration samples without need of containment devices like cuvettes, slides, chips, and accessories.
5. Auto-ranging multi pathlength technology allows for measurement of highly concentrated samples (e.g. up to 27,500 ng/μL dsDNA and 400 mg/mL IgG) and eliminates the need for sample dilutions.
6. Uses an embedded image sensor to provide digital image analysis monitoring for bubbles and broken sample columns.
7. Includes Auto-Blank and Auto-Measure options for increased efficiency.
8. Introduces Thermo Scientific™ Acclaro™ Sample Intelligence technology that can identify certain contaminants in nucleic acid and protein samples; reports corrected concentration of analyte.

In addition to the features described above, the Thermo Scientific NanoDrop OneC Spectrophotometer also offers:
1. An optional cuvette position for measuring dilute samples and performing kinetic experiments.
2. Ability to make cuvette measurements with the instrument arm up or down.

In the central nervous system, oligodendrocytes are cells responsible for producing myelin, the insulation around electrically active axons of neurons. Myelin facilitates fast electrical conductance in an analogous way as rubber insulation around electrical wiring. Proper development and maintenance of oligodendrocytes has important implications on demyelinating disease, including multiple sclerosis, a disease in which immune cells attack and destroy myelin, and leukodystrophies, childhood diseases in which myelin fails to form properly leading to progressive disability. In addition, recent studies also support a role for oligodendrocytes in psychiatric disease and in memory, including in Alzheimer’s disease.
Our lab focuses on understanding the cell biology and building blocks of the oligodendrocyte, and how they contribute to normal brain function and disease. The oligodendrocyte is a cell that is shaped like a tree, with many branches that terminate to form myelin sheaths, which wrap concentrically around the axon in many layers, much like a long paper towel roll or a bolt of fabric. Our questions primarily underly the structure of these branches – microtubules that form the roads of these cells – and the cargos that travel along them, including organelles and mRNA genetic messages.
