Background/Description of Requirement

The National Institute of Neurological Disorders and Stroke (NINDS) is a part of the National Institutes of Health (NIH), conducting research into the causes, treatment, and prevention of neurological disorders and stroke. PML is an opportunistic infection of the brain for which currently there is no cure.  The Experimental Immunotherapeutics Unit conducts a clinical research program dedicated to advancing our understanding of the pathophysiology of PML and the development of treatments.  To this end, there is an urgent need to improve our understanding of risk factors for this disease as well as development of biomarkers.  Magnetic resonance imaging is key for the diagnosis of the disease and for monitoring clinical course. However, in vivo imaging lacks histopathological specificity. One of the objectives of our group is detailed histopathological-radiological correlation studies. 10x Visium technology provides a way to investigate the molecular basis for magnetic resonance imaging findings, and to advance our understanding of the pathophysiology of this disease through acquisition of a spatial map of mRNA expression and organization of resident neuronal and glial cells and infiltrating immune cells from formalin fixed tissue.
