Background/Description of Requirement

The National Institute of Neurological Disorders and Stroke (NINDS), Division of Intramural Research, Laboratory of Neurobiology seeks to purchase a Purchase of a high-power Apple Mac Tower Pro computer as the main EM tomography-especially and cryo-EM tomography- computational resource for the lab. The work that we have done previously on EM tomography has been with freeze substituted using chemical fixatives with a trace of heavy metal, which gave sufficient contrast, this meant that we could analyze our tomograms with computer power we now have. But the computer power would not be sufficient dealing with cryo-EM data as much as we are asking for now, largely due to significantly larger datasets, computationally intense denoise the reconstruction algorithms and routines involved. So now that we are going to move to cloud microscopy, we need to add much more computing power and overall storage. Calculations and analyzing the tomographic data that is coming from cryo microscopy involves actually taking brain tissues and freezing them directly. And then open looking inside, with the electron beam, and then taking tomograms from the cropped areas in series of tilts. And we need over one hundred tilts for each tomogram since it is about data to be analyzed and then the data is collected analyzed and made into a tomogram, which allows us to study in three dimensions the structure of the proteins and special relationships among proteins at synapses in the brain
