Rationale/ Salient Characteristics
The darkfield illumination system and liquid light guide must have a Numerical Aperture of 1.2-1.4 and a focal distance of 1.5 mm. The field of view must be between 300 and 400 µm. The liquid light guide must have an inner diameter of 3mm and outer diameter of 5mm. The system must not weight more than 15 oz and be 98 x51 mm. in size.
Single-Sole Source Determination
Market research was conducted through google for darkfield condenser and only found one contractor whos product provides custom illuminator. By providing both critical and kohler illumination simultaneously, and in a manner that requires little to no alignment their illuminator gives a ~10x increase in darkfield signal, which is the key salient characteristics needed to support the research that the program needs to have. Background/Description of Requirement

The Cell Biology and Biophysics Unit (CBBU) at the National Institute of Neurological Disorders and Stroke (NINDS) studies microtubules(MTs) and microtubule associated proteins(MAPs) and their interactions. Tubulin is one of the most prevalent proteins in the brain and the polymerized form, microtubule’s are essential for a wide variety of cellular phenomena. Perturbation to the microtubule cytoskeleton can lead to sever degrative disease. The construction and disassembly of microtubules is a very regulated process in cells. To understand the fundamental principles we apply label free darkfield imaging to microtubule sample in vitro, CBBU is requesting the procurement of an Enhanced Darkfield Condenser and Liquid light guide. This will allow us to measure ten times more signal that our current darkfield setup. This increase in necessary to Image the degradation of the microtubule lattice and we can not currently make reliable measurements on existing equipment.
